丝氨酸衍生β-环糊精色谱柱的制备及色谱性能研究

摘要: 以本实验室合成的新型丝氨酸衍生β-环糊精键合硅胶固定相为研究对象,采用高压匀浆法装填高效液相色谱柱,对装填工艺进行了优化,获得良好的色谱柱效和装柱重现性.考察了衍生β-CD色谱柱分离位置异构体及手性化合物,并与未衍生β-CD色谱柱进行比较,讨论分离作用机理.结果表明,丝氨酸衍生β-环糊精对位置异构体以及含有苯环且具有极性基团的手性化合物具有很好的分离选择性.Abstract: Chromatography column packing of the newly prepared carbobenzyloxy-L-Ser derived β-CD bonded phase(SCDS),along with the underived β-CD bonded phase(CDS) were investigated.Satisfactory column efficiency,peak shape and reproducibility were obtained by optimizing high pressure column packing technology,such as packing pressure and slurry dispersing medium.The separation performance of SCDS for several position isomers and chiral compounds were also investigated and compared with CDS.The newly prepared column exhibited specific properties in realizing isomers separation.
